            Major problems with PCR-based clinical, environmental and forensic tests of samples containing blood and soil are false-negative results and low sensitivity caused by inhibitory substances. The effect of the main PCR inhibitors in blood and soil, hemoglobin and humic acid, is primarily associated with inactivation of Taq DNA polymerase. Therefore, various protocols and DNA extraction procedures are being used to purify DNA prior to PCR. These extra steps add to costs, are time-consuming, may not completely remove inhibitors, or may lead to losses of target DNA.
            As a novel alternative to these pre-PCR steps we have engineered Taq polymerase mutants, OmniTaq and Omni Klentaq, with high resistance to blood and soil inhibitors. They can tolerate up to 20-25% whole blood or serum in the reaction, or concentrations of soil which are completely inhibitory for other commercial Taq enzymes. And more good news - these enzymes can be used in real-time PCR applied directly with crude samples. The novel mutant Taq enzymes can eliminate, in many cases, the need to purify DNA prior to PCR and speed-up, lower the cost, and increase the efficiency of forensic DNA testing. With their unique features, OmniTaq and Omni Klentaq are perfectly suited for most reliable clinical and forensic applications.
